一種主要的醫(yī)院病原體，艱難梭菌（Clostridium difficile）引起抗生素相關(guān)性結(jié)腸炎和腸道炎癥性疾病，一起稱為CDI。
C艱難梭菌毒素如何引起腹瀉？ 通過(guò)釋放兩種大蛋白腸毒素，艱難梭菌毒素A（TcdA）和艱難梭菌毒素B（TcdB），艱難梭菌介導(dǎo)炎性腹瀉并損害腸??上皮細(xì)胞。施用于培養(yǎng)細(xì)胞時(shí)的毒素都會(huì)破壞細(xì)胞骨架并引發(fā)細(xì)胞凋亡依賴性細(xì)胞凋亡，盡管艱難梭菌毒素B的效力比艱難梭菌多很多倍。毒素A.兩種毒素還激活腸上皮細(xì)胞和免疫細(xì)胞以產(chǎn)生細(xì)胞因子和趨化因子，可能導(dǎo)致與嚴(yán)重CDI相關(guān)的并發(fā)癥。TcdA和TcdB的細(xì)胞內(nèi)靶標(biāo)是RhoGTP酶，其調(diào)節(jié)許多宿主細(xì)胞過(guò)程，包括建立上皮屏障和免疫細(xì)胞的遷移和信號(hào)傳導(dǎo)。當(dāng)Rho蛋白被艱難梭菌毒素A或B 糖基化時(shí)，信號(hào)被破壞，導(dǎo)致CDI的腸損傷和炎癥特征。
